Hb A2 is eluted from the column resin by a programmed buffer gradient of increasing ionic strength. Hb A2 measurement is best interpreted in conjunction with other testing in the Thalassemia Reflexive Panel and red cell indices and iron supply measurements.

- Interferences and Limitations
Variant hemoglobins such as Hb S, Hb C, Hb D interfere with measurement of Hb A2, so Hb A2 is not reported in the presence of these variants. Hb E co-elutes with Hb A2 and thus the level reported is the sum of percentages of Hb E and Hb A2.
