- Description
Initial testing is performed to qualitatively evaluate for the JAK2 V617F mutation. If this is negative, Calreticulin exon 9 mutation [CALR] testing will be reflexively ordered (with additional charge). If CALR screening is also found to be negative, the MPL sequencing by NGS [HCAPSG] will be reflexively ordered (with additional charge). This assay does not evaluate for other JAK2 mutations including exon 12 nor other exons. If indicated, JAK2 single gene sequencing by NGS (HCAPSG) is suggested for evaluation of these potential variants and should be separately ordered.

Interpretation

PCR amplification of JAK2 from genomic DNA, specific for V617F point mutation. May reflexively include allele specific PCR amplification of CALR and MPL exon 10 next-generation sequencing from genomic DNA.

Accepted:
Green Tops (Heparin), while green tops are accepted for testing, there is documentation that heparin can interfere with some PCR assays.Extracted DNA will only be accepted from CLIA certified laboratories.
Tissue from stained or cover-slipped slides
Unacceptable:
Specimens received beyond stability limit.
Specimens with low volume or inadequate WBC
Formalin-fixed paraffin embedded tissue (FFPE/PET)

- Deliver samples to Hematopathology as soon as possible.
- If sample is older than 48 hours, please store at 2-6°C upon receipt and send refrigerated.
- Samples stored at room temperature or refrigerated older than 7 days will be rejected.
- Frozen samples should be shipped on dry ice.

The stability limit for Blood and Bone Marrow is 7 days, samples beyond this limit will be rejected.
